Figure B.7 depicts the dideoxy method of DNA sequencing. This method was devised by Fred Sanger. The procedure is bipartite:(1)utilization of plasmid DNA and DNA polymerase to create a few hundred different pieces of DNA where each piece differs in length (from the other pieces)by one nucleotide; and (2)use of gel FIGURE B.7 Polyacrylamide gel containing bands of radioactive DNA. The figure shows a radioautographic image of a portion of a polyacrylamide gel that was used for determining a DNA sequence. Samples taken from four separate reactions described in the text, were applied to four wells, and these wells were named T, C, G, and A. The corresponding lanes containing the ladder pattern were also named T, C, G, and A. The order of application of the four samples is up to the discretion of the researcher, as this order may also be A-T-G-C, for example. Each band contains a homogeneous population of radioactive single-stranded DNA. All of the DNA molecules in the first lane begin with the primer sequence, and end in T. All of the DNA molecules in the second in the second lane begin with the primer sequence, and end in C, and so on. The figure depicts the image acquired after exposing the gel to the X-ray film for 1 day. This technique is called autoradiography, because the sample of interest (the DNA molecules embedded in the gel) are radioactive and provoke a graphic image in the film. The electric field, applied to the gel, forces the negatively charged molecules of DNA to migrate downward through the polyacrylamide gel. The separation of the DNA molecules from each other is based on their size, not charge. The polyacylamide gel acts as a sieve, alliwung smaller DNA molecules yo migrate faster through the gel matrix, and larger DNA molecules to migrate more slowly. The DNA molecules tend to stay in their own lanes, and tend not to cross-contaminate adjacent lanes.
