2.2. Extraction and determination of tea polyphenols

The polyphenols were isolated from fresh commercial

Chinese green tea, as described before. The tea was first refluxed with water. After filtration the tea liquid was concentrated in a rotary evaporator and then chloroform was added to remove caffeine,

lipids and chlorophyll. The aqueous phase was extracted

with-ethyl acetate and the ethyl acetate phase was freezedried.

The catechins content of the dried TP sample was determined

by HPLC with a DAD detector using a Hypersil

ODS C18 column (4.6 250 mm, 5 lm). The mobile phase

was a mixture of A (aqueous solution of 0.2% CH3CN) and

B (CH3OH). During the first 12 min, B was increased from

0% to 50%, and from 13 min to 20 min it was increased to

100%. The injection volume was 20 ll and the flow rate was

1.2 ml/min. Detection was set at 280 nm. Standard curves

of peak area, as a function of the concentrations of catechins,

were prepared for quantitative analysis.

2.3. Interaction of tea polyphenol with gelatin

Reactions took place in 10 ml volumetric flasks. TP was

dissolved in water to 0.05 mg/ml, 0.10 mg/ml and 0.15 mg/

ml. Gelatin solutions in water, whose concentrations varied

from 0.02 mg/ml to 0.16 mg/ml were prepared. TP and gelatin

solutions were mixed quantitatively in flasks under

shaking. After standing for 24 h at 25 C, the mixture

was centrifuged (MSE centrifuge, Model GF-8, 3000 rpm,

20 min) and the suspended substance (reaction products)

was removed. The supernatant was analyzed at 280 nm

by UV spectrophotometer (UV-2501PC, Japan). All reactions

were carried out in triplicate.

    2.2. 茶多酚 的 提煉 及 測定

    如前述,茶多酚是由新鮮商品綠茶所提煉出之萃取物。首先,將茶與水離析。過濾後將茶液減壓濃縮,再以氯仿去咖啡因、脂質、葉綠素。此時將該液體溶解於乙酸乙酯後將之冷凍乾燥。該茶多酚取樣中兒茶素之定量可經由安裝Hypersil ODS C18層析管(4.6 250 mm, 5 ml)之光二極體陣列式檢測器做高效液相層析而得。此一測定相為 A (濃度0.2%之氰化甲烷水溶液) 與 B (甲醇) 兩億體之混合物。測定過程前12分鐘將B從0%加至50%,於第13分鐘至第20分鐘加至100%。總注射(滴定)量為20毫升且流速為 1.2 毫升/分鐘。檢測定位設於280奈米。於定量分析時需準備峰區的標準曲線做為兒茶素濃縮的功用。

    2.3. 茶多酚 與 明膠 之 交互作用

    此反應可於10毫升級量瓶內發生。將茶多酚分別溶解至 0.05、0.10、及 0.15 (毫克/毫升)。再準備濃度為0.02 ~ 0.16 毫克/毫升 之明膠水溶液。而後,此兩種溶液定量的混合於量瓶內,均勻搖晃。於25*C(室溫)下靜置24小時後將其離心分離(MSE離心機, 型號: GF-8, 3000 轉/分, 作用長度20分鐘),之後將沉澱[or懸浮]物(反應生成物)移去。該上澄清液利用紫外線分光光度計 (型號UV-2501PC, 日本產)分析。所有反應均三份進行。

    我不是很明確其意故照字面翻, 但我再查若有結果會再來補充. 或者閣下對這一方面略之一二可以告知一下嗎?
