Topic- [Opposite effects of oxytocin on proliferation of osteosarcoma cell lines]

Introduction - The neurohypophyseal hormone and hypothalamic nonapeptide oxytocin has been found to induce several growth promoting effects.

Oxytocin promotes wound healing and weight gain in rats, and rat

pups which have received exogenous oxytocin during a short period

after birth, have a higherweight in adulthood.Women with high

oxytocin levels give birth to babies with a higher birth weight.

Oxytocin has insulin-like effects promoting uptake and oxidation of

glucose in adipocytes and stimulating lipogenesis. In addition,

oxytocin increases growth factors, and the development of

blastocysts as well as proliferation of several cells and cell lines

such as for example endothelial cells, adenohypophyseal cells,

adrenocortical cells, chondrocytes and myoblasts.

We have previously found that oxytocin increases proliferation of

human osteoblast-like cells and a human osteosarcoma cell line,

SaOS-2.

In contrast, oxytocin inhibits proliferation of endometrial- and

breast cancer cells, growth of certain brain tumours and proliferation

of some other osteosarcoma cell lines than SaOS-2.

Thus different effects of oxytocin have been reported depending on

cell type, and experimental conditions. A possible explanation behind

the opposite effects on proliferation of different cells is that the effect

of oxytocin may be dependent on the stage of cell differentiation.

Osteocalcin and alkaline phosphatase (ALP) are well-known osteoblasticmarkers.

Osteocalcin is themost abundant non-collagenous bone

matrix protein and is expressed late in osteoblast differentiation,

whereas ALP is a marker for early stage differentiated osteoblasts.

The human osteosarcoma cell line SaOS-2 is a relatively highly

differentiated cell line which expresses both ALP and osteocalcin.

TE-85 is another human osteosarcoma cell line which displays a

pre-osteoblastic phenotype with high proliferative capacity and low

expression of osteoblastic markers.
