Eachof the three trk receptors has preferential ligands

(NGF for TrkA, BDNF and NT-4 for TrkB, and NT-3 for TrkC),

and, in many instances, expression of the appropriate trk

receptor is required for a neurotrophin to elicit a biological

response (Barbacid, 1994; Snider, 1994).

Our knowledge of neurotrophin physiology derives mainly from studies performed with chickens and rodents(Lewin and Barde, 1996).

However, genes homologous to the mammalian neurotrophins

and their receptors have been cloned from members of all groups of vertebrates,

with the sole exception of elasmobranch fish (for review, see Barde, 1994).

The degree of amino acid sequence conservation differs widely among the different neurotrophins: NGF is significantly less conserved than BDNF or

NT-3 (Barde, 1994).

In addition, amphibian and mammalian neurons respond to human BDNF with comparable E50 (Cohen-Cory and Fraser, 1994).

These observations suggest that not only the amino acid sequence but also the

biological function of BDNF remained conserved during vertebrate evolution.

If this is the case, then homologous neurons of different vertebrates

are expected to present the same pattern of trkB expression.

The vertebrate retina has been a widely exploited system in comparative studies:Although the detailed morphology of single retinal neurons may vary considerably even among members of the same vertebrate group, and an

amazing variety of neuronal subtypes has been described

(see, e.g., Waessle and Boycott, 1991; Kolb, 1994).

All retinal neurons can be classified into five homologous classes,

and the laminar organization of the vertebrate retina is absolutely conserved

(for a comparative view of the retina, see Dowling, 1987).

The only retinal neurons presently known to respond to BDNF are retinal

ganglion cells (Johnson et al., 1986; Rodrı´guez-Tebar et al., 1989; Cohen-Cory and Fraser, 1994; Mansour-Roubaey et al., 1994).
